Description
Other Notes
BMT activator (also known as 5-Benzylmercapto-1H-tetrazole or BTT) demonstrates important advantages in the syntheses of oligonucleotides1. Coupling efficiencies of 99 % with impeccable quality2. Lower percentage of n-1 sequences3. Dramatic reduction of coupling times to under 3 minutes in RNA syntheses4. Efficient RNA synthesis using 50 % or less monomer5. Excellent batch to batch consistency for reproducible and stable oligo production
Application
A 0.3 M BMT solution (5-benzylmercaptotetrazole in anhydrous acetonitrile) is used as an activator reagent in solid-phase DNA and RNA oligonucleotide synthesis particularly during the phosphoramidite coupling step.
Purpose
Its purpose is to activate nucleoside phosphoramidites increasing their reactivity so they can efficiently couple with the free hydroxyl group of the growing oligonucleotide chain. This helps achieve effective and consistent chain elongation during synth
General Description
The solution contains 5-benzylmercaptotetrazole (BMT) at a concentration of 0.3 M dissolved in anhydrous acetonitrile. BMT functions as an acidic activator that facilitates the phosphoramidite coupling reaction while anhydrous acetonitrile provides a suitable moisture-free organic solvent.
