Description
Application
Oxidizer 0.05 M iodine in pyridinewater is used as an oxidation reagent during phosphoramidite-based synthesis of DNA and RNA oligonucleotides.
Purpose
The purpose of the oxidizer is to oxidize the trivalent phosphorus P(III) intermediate to the pentavalent phosphorus P(V) state thereby forming the normal phosphate backbone of the growing oligonucleotide.
General Description
The reagent is a solution containing approximately 0.05 M iodine (I) in a pyridinewater solvent system. Iodine acts as the oxidizing agent while pyridine provides a suitable basic reaction medium and water participates in the conversion of the phosphite intermediate to the phosphate linkage.
